Mechanotransduction current is essential for stability of the transducing stereocilia in mammalian auditory hair cells

چکیده
Mechanotransducer channels at the tips of sensory stereocilia of inner ear hair cells are gated by the tension of 'tip links' interconnecting stereocilia. To ensure maximal sensitivity, tip links are tensioned at rest, resulting in a continuous influx of Ca2+ into the cell. Here, we show that this constitutive Ca2+ influx, usually considered as potentially deleterious for hair cells, is in fact essential for stereocilia stability. In the auditory hair cells of young postnatal mice and rats, a reduction in mechanotransducer current, via pharmacological channel blockers or disruption of tip links, leads to stereocilia shape changes and shortening. These effects occur only in stereocilia that harbor mechanotransducer channels, recover upon blocker washout or tip link regeneration and can be replicated by manipulations of extracellular Ca2+ or intracellular Ca2+ buffering. Thus, our data provide the first experimental evidence for the dynamic control of stereocilia morphology by the mechanotransduction current.

Molecular Remodeling of Tip Links Underlies Mechanosensory Regeneration in Auditory Hair Cells
Sound detection by inner ear hair cells requires tip links that interconnect mechanosensory stereocilia and convey force to yet unidentified transduction channels. Current models postulate a static composition of the tip link, with protocadherin 15 (PCDH15) at the lower and cadherin 23 (CDH23) at the upper end of the link. In terminally differentiated mammalian auditory hair cells, tip links ar...
متن کاملThe murine catecholamine methyltransferase mTOMT is essential for mechanotransduction by cochlear hair cells
Hair cells of the cochlea are mechanosensors for the perception of sound. Mutations in the LRTOMT gene, which encodes a protein with homology to the catecholamine methyltransferase COMT that is linked to schizophrenia, cause deafness. Here, we show that Tomt/Comt2, the murine ortholog of LRTOMT, has an unexpected function in the regulation of mechanotransduction by hair cells. The role of mTOMT...
متن کاملThe organization of tip links and stereocilia on hair cells of bird and lizard basilar papillae.
Auditory papillae from three species of bird (pigeon, starling, and chick), and two species of European lizard (Podarcis muralis and Podarcis sicula) were examined by scanning electron microscopy. Hair bundles from all papillae showed tip links oriented along the direction of gradation in heights of the stereocilia (i.e. parallel to the hair-cell axis of bilateral symmetry, and so parallel to t...
متن کاملCalcium entry into stereocilia drives adaptation of the mechanoelectrical transducer current of mammalian cochlear hair cells.
Mechanotransduction in the auditory and vestibular systems depends on mechanosensitive ion channels in the stereociliary bundles that project from the apical surface of the sensory hair cells. In lower vertebrates, when the mechanoelectrical transducer (MET) channels are opened by movement of the bundle in the excitatory direction, Ca(2+) entry through the open MET channels causes adaptation, r...
متن کاملCadherin-23 May Be Dynamic in Hair Bundles of the Model Sea Anemone Nematostella vectensis
Cadherin 23 (CDH23), a component of tip links in hair cells of vertebrate animals, is essential to mechanotransduction by hair cells in the inner ear. A homolog of CDH23 occurs in hair bundles of sea anemones. Anemone hair bundles are located on the tentacles where they detect the swimming movements of nearby prey. The anemone CDH23 is predicted to be a large polypeptide featuring a short exopl...
